ABSTRACT | A method and assay system is provided for screening for antimicrobial agents that inhibit or otherwise disrupt the transcription of bacterial aminoacyl-tRNA synthetases, particularly those in Gram-positive bacteria. The method and assay utilizes purified RNA polymerase, isolated, for example, from either Bacillus subtilis or Escherichia coli, and a template DNA fragment which encodes glycyl-tRNA synthetase, containing the promoter and leader region of the B. subtilis glyQS gene, including the T-Box terminator/antiterminator. Incubation in the presence of nucleotide triphosphates results in synthesis of an RNA transcript initiating at the glyQS promoter and terminating at the leader region terminator. Read-through of the terminator, and synthesis of an extended transcript, is dependent on addition of purified glycyl-tRNA. The method and assay is dependent on the use of the specified tRNA species; non-specific tRNA fails to stimulate antitermination. The assay can be used to screen for inhibitors that specifically inhibit the T-box antitermination mechanism. |

ABSTRACT | Provided are four new fluorescent proteins. The proteins were derived from two wild-type fluorescent proteins: a red fluorescent protein (RFP) that was isolated from Actinodiscus or Discosoma sp. 1 and a green fluorescent protein (GFP) isolated from Montastraea cavernosa. Two mutant forms were generated from each wild-type protein. Each of the mutated forms has a higher fluorescence intensity than the respective wild-type form. The mutant forms of the fluorescent proteins allow for more sensitive detection of the fluorescence emitted by the proteins. Additionally, one of the mutant proteins is more resistant to photobleaching than its wild-type protein. The invention also encompasses isolated nucleic acids encoding the mutant forms of the wild-type RFP and GFP. |

ABSTRACT | Disclosed is a transformed yeast cell containing a first heterologous DNA sequence which codes for a mammalian G protein-coupled receptor and a second heterologous DNA sequence which codes for a mammalian G protein α subunit (mammalian Gα). The first and second heterologous DNA sequences are capable of expression in the cell, but the cell is incapable of expressing an endogenous G protein α-subunit (yeast Gα). The cells are useful for screening compounds which affect the rate of dissociation of Gα from Gβγ in a cell. Also disclosed is a novel DNA expression vector useful for making cells as described above. The vector contains a first segment comprising at least a fragment of the extreme amino-terminal coding sequence of a yeast G protein-coupled receptor. A second segment is positioned downstream from the first segment (and in correct reading frame therewith), with the second segment comprising a DNA sequence encoding a heterologous G protein-coupled receptor. |

ABSTRACT | In proteomic research, it is often necessary to screen a large number of polypeptides for the presence of stable structure. Described herein are methods (referred to as MALDI MS-HX and SUPREX) for measuring the stability of proteins in a rapid, high-throughput fashion. The method employs hydrogen exchange to estimate the stability of quantities of unpurified protein extracts, using matrix-assisted laser desorption/ionization (MALDI) mass spectrometry. A method of quantitatively determining the stability of a test protein under native conditions is disclosed. The method includes the steps (a) providing a test protein; (b) contacting the protein with an exchange buffer comprising a denaturant and deuterium, the exchange buffer having a denaturant concentration; (c) contacting the test protein with a mass spectrometry matrix medium; (d) determining a change in mass of the test protein by mass spectrometry; (e) varying the denaturant concentration of the exchange buffer; (f) repeating steps (a)-(e) a desired number of times; and (g) quantitatively determining protein stability based on the change in mass of the test protein as a function of denaturant concentration, whereby the stability of a test protein under native conditions is quantitatively determined. |

ABSTRACT | Systems and methods are presented for cell simulation and cell state prediction. For example, a cellular biochemical network intrinsic to a phenotype of a cell can be simulated by specifying its components and their interrelationships. The various interrelationships can be represented with one or more mathematical equations which can be solved to simulate a first state of the cell. The simulated network can then be perturbed, and the equations representing the perturbed network can be solved to simulate a second state of the cell which can then be compared to the first state, identifying the effect of such perturbation on the network, and thereby identifying one or more components as targets. Alternatively, components of a cell can be identified as targets for interaction with therapeutic agents based upon an analytical approach, in which a stable phenotype of a cell is specified and correlated to the state of the cell and the role of that cellular state to its operation. A cellular biochemical network believed intrinsic to that phenotype can then be specified, mathematically represented, and perturbed, and the equations representing the perturbed network solved, thereby identifying one or more components as targets. |

ABSTRACT | A template for assessing damaged areas on an object, such as a fan blade or other item, includes a set of marks indicating the maximum areas of damage allowed on the periphery of the object. A guide enables the template to be aligned in proper position against the object for accurate visual inspection of the damaged areas. For example, one or more blend guides are included to allow the inspector to mark a blend area around the damage to enable maintenance personnel to smooth sharp, jagged edges of the damaged areas, thereby relieving stress points and improving the aerodynamic efficiency of a blade in an engine. The template can include a magnifying portion that allows the user to see more detail on the object. Linear and/or curved measurement scales can also be included on the template. |

ABSTRACT | A microfluidic device comprises a matrix of rotary flow reactors. The microfluidic matrix device offers a solution to the “world-to-chip” interface problem by accomplishing two important goals simultaneously: an economy of scale in reagent consumption is achieved, while simultaneously minimizing pipetting steps. N2 independent assays can be performed with only 2N+1 pipetting steps, using a single aliquot of enzyme amortized over all reactors. The chip reduces labor relative to conventional fluid handling techniques by using an order of magnitude less pipetting steps, and reduces cost by consuming two to three orders of magnitude less reagents per reaction. A PCR format has immediate applications in medical diagnosis and gene testing. Beyond PCR, the microfluidic matrix chip provides a universal and flexible platform for biological and chemical assays requiring parsimonious use of precious reagents and highly automated processing. |

ABSTRACT | The invention includes an ion mobility spectrometer having a liquid filled drift chamber. The chamber has an ionization region partitioned from and an ion separation region by a reversible ion-migration block. An electrical field within the chamber allows ions to migrate toward the electrode collector. Passage of ions from the ionization region is triggered by reversing the block allowing ions to migrate into the ion separation region. The invention includes a method of ion mobility analysis in liquid phase. Ions are mobilized to migrate through a drift liquid and are detected at an end of a drift chamber. The invention also includes a method of generating ions in a sample. A sample containing molecules in a first solvent is introduced into a second solvent through a charged capillary where the electrically charged sample is electro-disperses to ionize the molecules. |

ABSTRACT | A polarization independent (PI) interferometer design that can be built from standard optical components is described. Based upon a Michelson interferometer, the PI interferometer uses a 50/50 splitter and Faraday Rotator Mirrors (FM's). The interferometer achieves good optical characteristics, such as high extinction ratio (ER) and low insertion loss (IL). Lack of polarization sensitivity reduces interferometer construction tolerances and cost, enhances performance and utility, and expands the scope of interferometric based devices. Such characteristics can be used to construct flexible, high performance, polarization insensitive, multi-rate, self-calibrating, optical DPSK receivers, power combiners, optical filters and interleavers, all-optical switches, and cascaded interferometers. Since polarization is not maintained in standard fiber optic networks, a PI-DPSK receiver allows for use of more sensitive DPSK communications over fiber, without need for costly polarization control hardware. Other applications of PI interferometers include optical CDMA, secure communications, optical coherence tomography (OCT), and temporal gratings with ultra-precise timing. |

ABSTRACT | A method for storage cell design evaluation provides accurate information about state changes in static storage cells. A wordline select pulse is propagated along the wordline select path of the test row to an output driver circuit, in order to test the clock and/or address timing of the row, so that variation of access timing, read stability and writeability with wordline strength/access voltage can be determined. An access detection cell holds the input of the output driver circuit until a simulated access operation activated by the wordline select pulse is complete. Multiple test rows may be cascaded among columns to provide a long delay line or ring oscillator for improved measurement resolution. |

ABSTRACT | The present invention is a method and system for estimating interference. Packet characteristics of relevant packets within the channel at a given point in time may be processed. For example, a power level, occupied frequency and corresponding time references may be accumulated and stored for each relevant packet within the channel at a given point in time. As each packet is processed for demodulation, the channel samples, at which interference occurred, may be appropriately weighted based on the packet characteristics. Advantageously, the nature of the interfering channel traffic may be measured to properly de-weight the interference in a computationally efficient manner. |

ABSTRACT | A method for estimating uncorrelated noise in a distributed record of data including data samples that represent both signal and noise, provided that the signal is substantially in phase between adjacent data samples. The method begins with dividing the record of data into an even number of equal intervals. The method proceeds compiling a reduced data record by subtracting from every other data sample in the record of data one data sample that is adjacent to it. Finally, the method entails a step of estimating the noise power in the signal relative to the signal power by calculating the standard deviation as a measure of the magnitude of the noise. |

ABSTRACT | A method and apparatus for producing virtual sound sources that are externally perceived and positioned at any orientation in azimuth and elevation from a listener is described. In this system, a set of speakers is mounted in a location near the temple of a listener's head. A head tracking system determines the location and orientation of the listeners head and provides the measurements to a computer which processes audio signals, from an audio source, in conjunction with a head related transfer function (HRTF) filter to produce spatialized audio. The HRTF filter maintains the virtual location of the audio signals/sound, thus allowing the listener to change locations and head orientation without degradation of the audio signal. The audio system of the present invention produces virtual sound sources that are externally perceived and positioned at any desired orientation in azimuth and elevation from the user. |

ABSTRACT | A computer system for controlling a nonlinear physical process. The computer system comprises a linear controller and a neural network. The linear controller receives a command signal for control of the nonlinear physical process and a measured output signal from the output of the nonlinear physical process. The linear controller generates a control signal based on the command signal, a measured output signal, and a fixed linear model for the process. The neural network receives the control signal from the linear controller and the measured output signal from the output of the nonlinear physical process. The neural network uses the measured output signal to modify the connection weights of the neural network. The neural network also generates a modified control signal supplied to the linear controller to iterate a fixed point solution for the modified control signal used to control the nonlinear physical process. |

ABSTRACT | An encoder employs a trellis coded quantization (TCQ) unit and a compression unit. The TCQ uses a set of polynomials that have been selected to maximize granular gain. The TCQ unit operates on a block of samples from a source. The compression unit compresses bit planes of the TCQ output, using parity check matrices of corresponding LDPC codes, to obtain corresponding syndromes. The parity check matrices are selected so their compression performance approaches close to the limit for Slepian-Wolf coding. A decoder employs a decoding unit and an estimation unit. The decoding unit decodes the syndromes using side information to produce an estimate for the TCQ output. The side information is correlated with the source. The estimation unit estimates the block of source samples using the estimated TCQ output and the side information. Trellis coded vector quantization may be used as an alternative to TCQ. |

ABSTRACT | A method and computer program product for performing minimum cost routing with network coding is presented. The method and system model a network as a directed graph. A cost per unit flow is associated with each link of the directed graph. A link capacity is associated with each link of the directed graph. A network code is then computed that sets up a routing connection that achieves an optimal cost using the cost per unit flow for each link of the directed graph and using the link capacity for each link of the directed graph. |

ABSTRACT | Parametric shape recognition is achieved through a decomposition of shapes into a hierarchy of subshapes ordered by their decreasing restrictions. Instances of each of the subshapes are individually located in the design shape and then reconstructed to form an instance of the entire shape. The basis for the hierarchy of subshapes can be specified by the designer or based on the default parameter relations that come from architectural and engineering knowledge. The levels of the hierarchy are defined so that the most constrained lines of a shape are those lines that the designer intended exactly. These most constrained lines have specified parametric relations to other line segments and those relations, if altered, will compromise the designer's intentions. Conversely, the lowest level of the hierarchy, which contains the least constrained line segments, only implies a specific connectivity between line segments, necessitating a vaster search. The parametric recognition of curved line shapes used a two-step approach that first performs shape matching with an equivalent straight-line shape then checks those transformations for matching with the actual curved lines. This approach has advantages characteristic polygons as well as emergent shapes. |

ABSTRACT | Disclosed herein is a new method for the solid phase peptide synthesis (SPPS) of C-terminal peptide α thioesters using Fmoc/t-Bu chemistry. This method is based on the use of an aryl hydrazine linker, which is totally stable to conditions required for Fmoc-SPPS. When the peptide synthesis has been completed, activation of the linker is achieved by mild oxidation. The oxidation step converts the acyl-hydrazine group into a highly reactive acyl-diazene intermediate which reacts with an α-amino acid alkylthioester (H-AA-SR) to yield the corresponding peptide α-thioester in good yield. A variety of peptide thioesters, cyclic peptides and a fully functional Src homology 3 (SH3) protein domain have been successfully prepared. |

ABSTRACT | A method and apparatus for performing simultaneous multi-point measurements of multiple velocity components in a gas flow is described. Pulses of laser light are directed to a measurement region of unseeded gas to produce Rayleigh or Mie scattered light in a plurality of directions. The Rayleigh or Mie scattered light is collected from multiple directions and combined in a single collimated light beam. The Rayleigh or Mie scattered light is then mixed together with a reference laser light before it is passed through a single planar Fabry-Perot interferometer for spectral analysis. At the output of the interferometer, a high-sensitivity CCD camera images the interference fringe pattern. This pattern contains the spectral and spatial information from both the Rayleigh scattered light and the reference laser light. Interferogram processing software extracts and analyzes spectral profiles to determine the velocity components of the gas flow at multiple points in the measurement region. The Rayleigh light rejected by the interferometer is recirculated to increase the accuracy and the applicability of the method for measurements at high temperatures without requiring an increase in the laser energy. |

ABSTRACT | A low-frequency, temporally asymmetric watermark carrier decreases in luminance at a slower rate than it increases in luminance. This allows the carrier to be embedded, e.g., in a video work, in a manner that renders the resultant watermark virtually invisible to the human eye. Moreover, the resultant watermark is substantially resistant to common pirating techniques in which high-frequency visual content (e.g., including watermark data) tends to be filtered out by piracy processing. |

ABSTRACT | Systems, methods, and software consistent with the present invention use combinations of two or more letters (xgraphs) or characters, such as three letter combinations (trigraphs), to aid in deciphering ambiguous characters in OCR systems. Embodiments consistent with the invention compare possible xgraphs constructed by substitution for the ambiguous OCR-read character(s) to a reference set of xgraphs. The reference set of xgraphs contains predetermined valid xgraphs, which may be ranked by preference. If a possible xgraph matches a reference xgraph, then the ambiguous character(s) are resolved in favor of the characters in the reference xgraph. Embodiments may also use the context of the ambiguous character string to aid in resolving the ambiguity where more than one possible solution exists. |

How To Use This Page
THE FEDINVENT PATENT DETAILS PAGE
Each week, FedInvent analyzes newly granted patents and published patent applications whose origins lead back to funding by the US Federal Government. The FedInvent Patent Details page is a companion to the weekly FedInvents Patents Report.
This week's information is published in the FedInvent Patents report for Tuesday, August 19, 2008.
The FedInvent Weekly Patent Details Page contains a subset of patent information to provide a deeper dive into the week’s taxpayer-funded patents to help the reader better understand where a patent fits in the federal innovation ecosphere.
HOW IS THE INFORMATION ORGANIZED?
Patents are organized by the funding agency. Within each group, the patents are organized in numeric order. A patent funded by more than one agency will appear in the section of each of the agencies that funded the research and development that resulted in the invention. This approach gives the reader a complete view of the department or agency activity for the week.
WHAT INFORMATION WILL I FIND?
THE PANEL
There is a panel for each patent that contains the patent number and the title of the patent. When you click the panel, it opens to reveal the following information:
FUNDED BY
The agencies that funded the grants, contracts, or other research agreements that resulted in the patent. FedInvent includes as much information on the source of the funding as possible. The information is presented in a hierarchy going from the Federal Department down to the agencies, subagencies, and offices that funded the work. Here are two examples:
Department of Health and Human Services (HHS)
National Institutes of Health (NIH)
National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK)
Department of Defense (DOD)
Defense Advanced Research Projects Agency (DARPA)
Army Research Office (ARO)
We do our best to provide detailed information about the funding. In some cases, the patent only reports limited information on the origins of the funding. FedInvents presents what it can confirm. We add the patents without the information required by the Bayh-Dole Act to our list of patents worthy of further investigation.
APPLICANT(S) and ASSIGNEES
FedInvent includes both the Applicants and the Assignees because having both provides more information about where the inventive work was done and by what organizations. Many organizations — universities, corporations, and federal agencies — standardize the Assignee/Owner information by the time a patent is granted. In the case of federal patents, many of the patents use the agency headquarters information for patent assignment.
Showing just the headquarters address would make Washington, DC the epicenter of all taxpayer-funded research and development. Providing both the applicant information and the assignee information provides a more accurate picture of where important taxpayer funded innovation is happening in America. Here are two examples from two different patents:
APPLICANT: U.S. Army Research Laboratory, Adelphi, MD
ASSIGNEE: The United States of America as represented by the Secretary of the Army Washington, DC
APPLICANT: Optech Ventures, LLC (Torrance, California)
ASSIGNEE(S): The Regents of the University of California (Oakland, California); Optech Ventures, LLC (Torrance, California)
INVENTOR(S)
The inventors appear in the same order as they appear on the patent. FedInvents presents the names in first name/last name order because they are easier to read than the last name/first name order of the names on the USPTO patent documents.
ABSTRACT
The abstract as it appears on the patent.
FILED
The date of the patent application including the day of the week.
APPL NO
This is the patent application serial number. If you’d like to learn more about how application serial numbers work you can go to the Lists Page.
ART UNIT
Patent data includes the Art Unit where a patent was examined. (The Art Unit isn’t available for published patent applications.) The Art Unit provides insight into what group of patent examiners prosecuted the patent application and the subject matter that the examiners work on. For example:
3793 — Medical Instruments, Diagnostic Equipment, and Treatment Devices
You can learn more about ART UNITS on the FedInvent Patents Weekly panel called About Tech Center or you can find information on the FedInvent Lists Page.
CURRENT CPC
Current CPC provides a list of the Cooperative Patent Classification symbols assigned to the patent. These are the CPC symbols assigned at the time the patent was granted.
The FedInvent Project is a patent classification maximalist endeavor or put another way, we believe that more you understand about patent classification the more you'll learn about the nature of the invention and the types of work that the federal government is funding.
The symbol presented in BOLD is the symbol identified as the "first" classification which is the most relevant classification on the patent. The date that follows the symbol is the date of the most recent revision to the art classed there.
- A61B 1/149 (20130101)
- A61B 1/71 (20130101)
- A61B 1/105 (20130101)
The CPC symbols match the classifications found on the PDF version of the patent. Over time, the classifications on the full-text version of the patent change to reflect how USPTO organizes patent art to support its examiners. The two sets of CPCs don’t always match.
VIEW PATENT
As of June 2021, we include two ways to view a patent at USPTO. FedInvent provides a link to the Full-Text Version of the patent and a link to the PDF version of the patent.
